There are now many reports of the successful design of high affinity low molecular weight ligands to bind the active sites of enzymes. Inhibition of certain enzyme activites can lead to treatment for infectious or metabolic diseases, but the designed inhibitors, however successful they may be in biochemical assays, often fail the test of efficacy in animals.
