Objective The experiment was designed to research a detection method for hepatitis B surface antigen (HBsAg) with wide application and high sensitivity. Method On the base of antibody reacting specially with HBsAg, biotinylated antibody and avidin-DNA conjugate were added in step by step. The relevant antigen was indicated by amplification and detection of DNA. Result The sensitivity of immuno-PCR was 300 times as sensitive as that of conventional ELISA. Conclusion Immuno-PCR was a very sensitive detection method for HBsAg.%目的:探讨一种适用性强、敏感性高的乙型肝炎(乙肝)表面抗原检测方法。方法:在乙肝表面抗原与相应抗体特异性反应的基础上,逐步加入生物素化抗体、亲合素-DM耦联物,PCR扩增相应的DNA标记,通过检测DNA来反映相应抗原的量。结果:免疫PCR检测敏感度为常规酶联免疫试剂盒的300倍。结论:免疫PCR是一种适用于乙肝表面抗原的、高度敏感的检测方法。
