摘要： Tip60 is a specific member of MYST (Moz-Ybf2/Sas3-Sas2-Tip60) family of nuclear histone acetyltransferases (HAT). It is essential for cellular survival, differentiation, and metabolism. A putative canonical NLS motif between the chromo domain and the zinc finger of Tip60 was identified. Here we show evidence that Tip60 is associated with importin α as its substrate and transported from cytoplasm to the nucleus. Pull down assay revealed that Tip60 was physically associated with importin α both in vivo and in vitro. Confocal microscopic observation showed that Tip60 and importin α were co-localized with each other. The localization of Tip60 to the nuclear and its interaction with importin α was disrupted when its putative NLS motif for binding to importin α was mutated (219RKRK222 → 219AAAA222). However, attachment of this putative NLS motif to a cytoplasmic protein (YAP 1-210 fragment) promoted its nuclear localization. Based on transient transfection, Tip60 NLS motif mutant showed a substantial reduction in self-acetylation, HAT activity, and apoptotic ability whereas wild type Tip60 did not show such reduction. Taken together, our results demonstrate that importin α transports Tip60 from the cytoplasm to the nucleus through binding to the putative NLS motif of Tip60 for its tumor suppressing function.

摘要： Lysine acetylation of histone plays an important role in regulation of gene expression in plant cells ,meanwhile it affects the enzyme activity of glycolysis ,tricarboxylic acid cycle and photosynthetic pathway. Therefore ,it is of great significance to identify histone acetyltransferase(HAT) and histone deacetylase(HDAC) of metabolism regulating enzymes. On the contrary ,metabolites such as acetyl-CoA and NAD+are involved in processes of protein acetylation and deacetylation ,their intracellular levels may influence on activity of HAT and HDAC. This paper illustrates the role of HAT ,HDAC and their research progress from protein lysine acetylation process ,classification and cell distribution ,and metabolic enzyme activity.%植物细胞中组蛋白赖氨酸乙酰化修饰对基因表达具有重要的作用，同时影响糖酵解、三羧酸循环和光合作用途径中非组蛋白的代谢酶活性。因此，识别参与调控代谢酶活性的HAT和HDAC具有重要意义。相反，代谢物如乙酰-CoA和NAD+参与蛋白可逆乙酰化进程，并且它们在细胞内的水平可能影响组蛋白乙酰化酶和组蛋白去乙酰化酶的活性。从组蛋白质赖氨酸乙酰化可逆过程、蛋白乙酰化酶和组蛋白去乙酰化酶分类及其细胞内分布和代谢酶的活性研究阐述了HAT和HDAC的作用及其研究进展。

摘要： 目的探讨稀释式自身输血（HAT）在外科手术应用中的有效性和可行性。方法：选择使用HAT的60例外科择期手术病例为治疗组；使用异体输血法的70例外科手术病例为对照组。记录2组患者术中接受不同输血治疗的情况，对其输血前后的免疫功能指标和血常规指标变化进行统计分析。结果：HAT能避免和减少输用异体血液；治疗组无输血不良反应；免疫功能指标和免疫球蛋白变化：在治疗组输血前后差异无显著性（P〉0．05）；对照组的差异有显著性（P〈0．05）；两组组间比较差异有显著性（P〈0．05）。血常规指标变化两组间差异无显著性（P〉0．05）。结论：HAT应用于外科手术中是安全有效和可行的，值得推广应用。

摘要： We use the general form of hat matrix and DFBETA measures to detect the influential observations in order to estimate the Divisia price index number when the error structure is first order serial correlation. An example is presented with reference to price data of Pakistan. Hat values show the noteworthy findings that the corresponding weights of consumer items have large influence on the parameter estimates and are not affected by the parameter of autoregressive process AR(1). Whereas DFBETAs for Divisia index numbers depend on both the weights and autoregressive parameter.

摘要： This paper, on the first hand, deals with the problem of estimation of Laspeyre price index number when the errors are assumed to be generated from AR(2) process. The general expression of hat matrix and DFBETA measure to find the influential consumer commodities in stochastic Laspeyre price model with AR(2) errors are developed on the other. The hat values show the noteworthy findings that the corresponding weights of consumer items have large influence on the parameter estimates for simple Laspeyre price index number and are not affected by the parameter of autoregressive process of order two. While, DFBETA measures are the functions of both weights and autocorrelation parameters. Lastly, an example is presented with reference to price data of Pakistan, and shows its practical importance in financial time series.

摘要： [目的]构建小鼠p300-HAT的真核表达质粒.[方法]利用PCR技术扩增p300的HAT域,然后构建p300的HAT域的真核表达质粒(p300-HAT-EGFP),并通过PCR验证和酶切验证重组质粒的正确性;提取不含内毒素的质粒,转染HepG2细胞,通过观测绿色荧光检测转染效率.[结果]PCR产物的电泳结果显示,成功扩增了小鼠p300基因HAT结构域,构建了小鼠p300基因HAT结构域的真核表达质粒,将其命名为m-p300-HAT-pEGFP-C1.重组质粒可以成功表达融合绿色荧光蛋白目的基因蛋白.[结论]p300-HAT-EGFP重组质粒的成功构建为后续开展巨噬细胞NF-kB信号通路的研究奠定了基础.

摘要： Objective To observe clinical effect of exposure therapy after hair apposition technique (HAT).Methods A total of 86 patients with simple scalp laceration in our emergency department during January 2009 and November 2012 were randomly divided into control group (n =43) and observation group (n =43),Patients in the two groups were treated with thorough debridement and HAT after disinfection.The wounds were covered by accessories in control group,while in observation group the wounds were exposed.Effective rate,healing time and visual analogue scales (VAS) of the two groups were observed and compared.Results In observation group,the healing rate and effective rate were 95.3％ and 97.7 ％ respectively,which were significantly higher than 79.1％ and 86％ in observation group (P ＜0.05),postoperative VAS score in observation group was significantly lower than that in control group (P＜0.05) ; healing time of wound were (4.5 ± 1.32) d in observation group and (6.7 ± 2.15) d in control group,and the difference was statistically significant (P ＜ 0.05).Conclusion HAT combined with exposure therapy in treatment of simple scalp laceration may have significant effect compared with covered dressings method.%目的 观察头发交叉固定术(hair apposition technique,HAT)处理后采用暴露疗法的临床效果.方法 选择2009年1月-2012年11月我院急诊科收治的单纯头皮裂伤86例,随机分为观察组与对照组,每组43例.2组均彻底清创、消毒后行HAT,观察组暴露伤口,对照组辅料覆盖伤口.观察比较两组有效率、痊愈时间和视觉模拟评分法(visual analogue scole,VAS)评分.结果 观察组痊愈率95.3％,有效率97.7％,均显著高于对照组的79.1％和86％ (P ＜0.05),观察组术后VAS评分高于对照组(P＞0.05)；伤口愈合时间,观察组(4.5±1.32)d,对照组(6.7±2.15)d,差异有统计学意义(P＜0.05).结论 单纯头皮裂伤后应用HAT并暴露疗法疗效优于覆盖敷料法,有临床应用价值.

摘要： The S-Nitrosylation of protein thiol groups by NO is a widely recognized protein modification. The treat-ment of cells with NOBF4 induces the S-nitrosylation of FE65. In this study, we present evidence showing that FE65 modified by NO (Nitric Oxide) via S-nitrosylation induces functional changes in the protein that inhibits the HAT activity of Tip60. The results of mutational analysis of FE65 demonstrated further that the cysteine residue of FE65 (Cys440) is critical to the process of S-nitrosylation. The mutation of the cysteine residue which completely ablated the S-nitrosylation of FE65 also lost its inhibitory effects on Tip60 HAT activity. Thus, our findings show, for the first time, that the novel regulation mechanism of Tip60 activity may operate via FE65 binding, which is enhanced by S-nitrosylation on the FE65 Cys440 residue. This study describes the interaction between FE65 and Tip60, which is enhanced by a posttransla-tional modification of FE65 (through S-nitrosylation) by NO, promoting the association of the FE65-Tip60 protein complex and inhibiting both the HAT activity of Tip60 and cell death.

摘要： 本发明涉及分子标记技术领域，特别涉及基于油茶转录组hAT转座子的分子标记引物组及其应用，本发明的引物是针对油茶转录组hAT转座子设计的，设计出来的7对引物具有较高的多态信息含量(PIC)，有良好的多态性，能对油茶品种进行良好区分，本申请印证了利用普通油茶开发hAT转座子标记的可行性，并对普通油茶、越南油茶、香花油茶三个品种的油茶进行遗传多样性分析，不仅能为油茶提供新颖优质的分子标记，也为油茶的分子改良奠定良好基础可为今后开展遗传育种、基因组作图、基因定位和物种亲缘关系鉴别等研究提供新引物。

摘要： 本发明公开了一种小檗碱衍生物及其制备方法和其作为p300 HAT小分子抑制剂的应用，属于药物化学技术领域，本发明以天然产物黄连中的有效成分盐酸小檗碱为研究对象，对其进行结构修饰与改造后，获得了一系列盐酸小檗碱衍生物。该类小檗碱衍生物对p300 HAT具有高活性、高选择性和安全性高的特点，解决了现有p300 HAT小分子抑制剂中存在的细胞毒性高、亲和力弱、活性低和选择性差的问题。

摘要： 本发明公开了一种单克隆抗体制备用HAT半固体筛选培养基，涉及生物技术领域。本发明在提取单克隆抗体制备用HAT半固体筛选培养基时，先将生物组织进行粉磨，脱脂，酶解制得生物蛋白水解物，再将石墨烯改性后自身交联反应形成沉积膜，将沉积膜与青霉素和链霉素反应制得载体膜，将蛋白水解物，胎牛血清和聚乙二醇混合沉积在载体膜上，制得单克隆抗体制备用HAT半固体筛选培养基。本发明制备的单克隆抗体制备用HAT半固体筛选培养基具有良好的杂瘤细胞培养效果。
